RESUMO

Accurate specification of the drugs' solubility is known as an important activity to appropriately manage the supercritical impregnation process. Over the last decades, the application of supercritical fluids (SCFs), mainly CO2, has found great interest as a promising solution to dominate the limitations of traditional methods including high toxicity, difficulty of control, high expense and low stability. Oxaprozin is an efficient off-patent nonsteroidal anti-inflammatory drug (NSAID), which is being extensively used for the pain management of patients suffering from chronic musculoskeletal disorders such as rheumatoid arthritis. In this paper, the prominent purpose of the authors is to predict and consequently optimize the solubility of Oxaprozin inside the CO2SCF. To do this, the authors employed two basic models and improved them with the Adaboost ensemble method. The base models include Gaussian process regression (GPR) and decision tree (DT). We optimized and evaluated the hyper-parameters of them using standard metrics. Boosted DT has an MAE error rate, an R2-score, and an MAPE of 6.806E-05, 0.980, and 4.511E-01, respectively. Also, boosted GPR has an R2-score of 0.998 and its MAPE error is 3.929E-02, and with MAE it has an error rate of 5.024E-06. So, boosted GPR was chosen as the best model, and the best values were: (T = 3.38E + 02, P = 4.0E + 02, Solubility = 0.001241).

341 entries comprising of 250 genotypes/lines and 91 gene differentials were tested for leaf rust (Puccinia triticina Erik) in different ecological zones of Punjab during 2016-17 and 2017-18. Each entry was planted in a single 1 m long row and Morocco was used as a spreader. Data on leaf rust severity was recorded once in 3rd week of March during both study years at all locations by following Modified Cobb Scale while the data was recorded three times on 2nd, 22nd and 29th March during 2018 at Faisalabad location to study rust development pattern. The disease severity ranged from 0-100S during 2016-17 and from 0-80S during 2017-18. The genotype HYT 60-5 and the genes Lr-19, Lr-26 and Lr 27+31 showed no disease symptoms at any location during both the study years. These genes can be used for future breeding material development. Area under disease progressive curve (AUDPC), calculated on the basis of periodical readings from Faisalabad, ranged from 0-550 and the susceptible check Morocco has AUDPC value of 600. 120 entries including HYT 60-5 have disease progression 0, which showed that there may be a major gene based resistance in these entries. Area under disease progressive curve/Day (AUDPC/DAY) was calculated for the rest of 130 genotypes to have an understanding of the disease progression pattern and out of which 43 entries have AUDPC/Day value ranging from 1-2 and 28 entries have AUDPC/Day value ranging from 2-3 which revealed that these entries are very useful for use in breeding for durable rust resistance and can be utilized as a parent in back cross and top cross breeding schemes. Material with AUDPC value less than 10 is the best source of resistance against the leaf rust. Varieties/advanced lines, Ujala-16, V-14154, and V-14124 have shown slow rust development and are very good sources of resistance. Similarly, HYT 60-5 has proven an excellent source of resistance. The advance line V-14154 has been approved as a commercial cultivar by the name "Akbar-19".

In our study, a unique ratiometric fluorescent sensor for the rapid detection of arginine (Arg) and acetaminophen (AP) was constructed by the integration of blue fluorescent N-CDs and yellowish-green fluorescent calcein. The N-CD/calcein ratiometric fluorescent sensor exhibited dual emission at 435 and 519 nm under the same excitation wavelength of 370 nm, and caused potential Förster resonance energy transfer (FRET) from N-CDs to calcein. When detecting Arg, the blue fluorescence from the N-CDs of the N-CD/calcein sensor was quenched by the interaction of N-CDs and Arg. Then, the fluorescence of our sensor was recovered with the addition of AP, possibly due to the stronger association between AP and Arg, leading to the dissociation of Arg from N-CDs. Meanwhile, we observed an obvious fluorescence change from blue to green, then back to blue, when Arg and AP were added, exhibiting the "on-off-on" pattern. Next, we determined the detection limits of the N-CD/calcein sensor to Arg and AP, which were as low as 0.08 µM and 0.02 µM, respectively. Furthermore, we discovered that the fluorescence changes of the N-CD/calcein sensor were only responsible for Arg and AP. These results suggested its high sensitivity and specificity for Arg and AP detection. In addition, we have successfully achieved its application in bovine serum samples, indicating its practicality. Lastly, the logic gate was generated by the N-CD/calcein sensor and presented its good reversibility. Overall, we have demonstrated that our N-CD/calcein sensor is a powerful sensor to detect Arg and AP and that it has potential applications in biological analysis and imaging.

Eucalyptus wood is made up of lignocellulosic material; this lignocellulosic material contains two types of biopolymers, i.e., carbohydrate and aromatic polymers. In this study, this lignocellulosic material was used to prepare biochar. Three biochar, i.e., laboratory-based (B1), barrel-based (B2), and brick kiln-biochar (B3), were used for fluoride and arsenic removal from aqueous solution. Barrel-based biochar was prepared by using the two-barrel method's alteration. The highest fluoride removal (99%) was attained at pH 2 in the presence of B1, while in the presence of B2 and B3, maximum fluoride removal was 90% and 45.7%, respectively. At pH 10, the maximum arsenic removal in the presence of B1, B2, and B3 was 96%, 94%, and 93%, respectively. The surface characteristics obtained by Fourier-transform infrared spectroscopy (FTIR) showed the presence of carbonyl group (C-O), and alkene (C=C) functional groups on all the three studied biochars. Isotherm studies showed that the adsorption was monolayered (all the adsorbed molecules were in contact with the surface layer of the adsorbent) as the Langmuir isotherm model best fits the obtained data. Adsorption kinetics was also performed. The R2 value supports the pseudo-second-order kinetics, which means that chemisorption was involved in adsorbing fluoride and arsenic. It is concluded that B1 gives maximum removal for both fluoride (99%) and arsenic (96%). The study shows that lignocellulose-based biochar can be used for arsenic and fluoride removal from water.

In current study, analysis is presented for peristaltic motion of applied magnetic field and entropy generation within couple stress (Cu/H2O) nanofluid through an endoscope. An endoscope contains two coaxial cylindrical tubes in which the internal tube is nonflexible while the external tube has sinusoidal wave passing through the boundary. Influences of mixed convection along with applied magnetic field are encountered as well. Formulated governing model is fabricated introducing long wavelength and creeping Stokesian flow approximation which are then analyzed numerically by utilizing Adams Bashforth method. For a physical insight, results are demonstrated to examine the behaviors of flow profiles and entropy generation number for emerging flow parameters with the help of graphs, bar-charts and tables.

BACKGROUND: Testicular injury is one of the most serious problems associated with diabetes mellitus. The present study aimed to compare the effects of two different doses of nobiletin and analyze its mechanisms of action against diabetes-induced testicular impairment in rats. METHODS AND RESULTS: Streptozotocin injection was used to induce diabetes. Diabetic rats received nobiletin orally at 10 or 25 mg/kg daily for 30 days. Diabetic rats displayed significant elevations in glucose, glycosylated hemoglobin (HbA1c), Homeostatic Model of Insulin Resistance (HOMA-IR), and pro-inflammatory cytokines, while the serum levels of insulin, testosterone, luteinizing hormone (LH), and follicle-stimulating hormone (FSH) were significantly reduced. Histological changes to positivity for caspase-3 and decreased androgen receptors (AR) immunoexpression were observed in diabetic rats. Both doses of nobiletin improved hyperglycemia, reduced pro-inflammatory cytokines, and augmented insulin, testosterone, LH, and FSH levels. LH and FSH receptors and cytochrome P450 17 α-hydroxylase (CYP17A1) were markedly downregulated in terms of both gene and protein expression in testicular tissues of the diabetic group, effects that were markedly ameliorated with both doses of nobiletin. In addition, both doses significantly reduced lipid peroxidation and caspase-3 immunoexpression and improved the activity of the antioxidant enzymes and AR in testicular tissues of the diabetic group. CONCLUSION: Both nobiletin doses showed protective effects against diabetes-induced testicular injury by reducing oxidative stress, hyperglycemia, inflammation, and caspase-3 and upregulating the hypophysis-gonadal axis and AR. The high dose of nobiletin was more effective than the lower one.

Biodiesel commercialization is questionable due to poor brake thermal efficiency. Biodiesel utilization should be improved with the addition of fuel additives. Hydrogen peroxide is a potential fuel additive due to extra hydrogen and oxygen content, which improves the combustion process. In this experimental study, biodiesel has been produced from Jatropha oil employing catalyzed transesterification homogeneously to examine its influence on the performance and emissions at engine loads with 1500 rpm utilizing a four-stroke single-cylinder diesel engine. D60B40 (having 60% diesel and 40% biodiesel) and D60B30A10 (60% diesel, 30% biodiesel and 10% hydrogen peroxide (H2O2)), are the fuel mixtures in the current study. The addition of H2O2 reduces emissions and enhances the combustion process. This effect occurred due to the micro-explosion of the injected fuel particles (which increases in-cylinder pressure and heat release rate (HRR)). An increase of 20% in BTE and 25% reduction in BSFC for D60B30A10 was observed compared to D60B40. Significant reduction in emissions of HC up to 17.54%, smoke by 24.6% CO2 by 3.53%, and an increase in NOx was noticed when the engine is operated with D60B30A10. The HRR increased up to 18.6%, ID reduced by 10.82%, and in-cylinder pressure increased by 8.5%. Test runs can be minimized as per Taguchi's design of experiments. It is possible to provide the estimates for the full factorial design of experiments. Exhaust gas temperature standards are evaluated and examined for all fuel blends.

Prandtl-Eyring hybrid nanofluid (P-EHNF) heat transfer and entropy generation were studied in this article. A slippery heated surface is used to test the flow and thermal transport properties of P-EHNF nanofluid. This investigation will also examine the effects of nano solid tubes morphologies, porosity materials, Cattaneo-Christov heat flow, and radiative flux. Predominant flow equations are written as partial differential equations (PDE). To find the solution, the PDEs were transformed into ordinary differential equations (ODEs), then the Keller box numerical approach was used to solve the ODEs. Single-walled carbon nanotubes (SWCNT) and multi-walled carbon nanotubes (MWCNT) using Engine Oil (EO) as a base fluid are studied in this work. The flow, temperature, drag force, Nusselt amount, and entropy measurement visually show significant findings for various variables. Notably, the comparison of P-EHNF's (MWCNT-SWCNT/EO) heat transfer rate with conventional nanofluid (SWCNT-EO) results in ever more significant upsurges. Spherical-shaped nano solid particles have the highest heat transport, whereas lamina-shaped nano solid particles exhibit the lowest heat transport. The model's entropy increases as the size of the nanoparticles get larger. A similar effect is seen when the radiative flow and the Prandtl-Eyring variable-II are improved.

Survey of literature unveils that nanofluids are more efficient for heat transport in comparison to the traditional fluids. However, the enlightenment of developed techniques for the augmentation of heat transport in nanomaterials has considerable gaps and, consequently, an extensive investigation for aforementioned models is vital. The ongoing investigation aims to study the 2-D, incompressible Jeffrey nanofluid heat transference flow due to a stretchable surface. Furthermore, the effect of dispersion of graphene nanoparticles in base liquid ethylene glycol (EG) on the performance of flow and heat transport using the Tawari-Das model in the existence of Ohmic heating (electroconductive heating) and viscous heat dissipation is contemplated. The boundary-layer PDEs are reconstituted as ODEs employing appropriate similarity transformation. Keller-Box Method (KBM) is utilized to determine the numerical findings of the problem. Graphene conducts heat greater in rate than all of the other materials and it is a good conductor of electrical energy. Graphene/EG nanofluid is employed to look out the parametric aspects of heat transport flow, drag coefficient, and heat transference rate phenomena with the aid of graphs and tables. The numerical outcomes indicate that concentration and magnetic field abate the shear stresses for the nanofluid. An increase of Graphene nanoparticle volume fraction parameter can boost the heat transport rate. The effect of Prandtl Number is to slow down the rate of heat transport as well as decelerate the temperature. Additionally, the rate of heat transportation augments on a surface under Deborah's number. Results indicate that the temperature of the graphene-EG nanofluid is greater than the convectional fluid hence graphene-EG nanofluid gets more important in the cooling process, biosensors and drug delivery than conventional fluids.

Air-frying is an innovative technique for food frying that uses hot air circulation to prepare healthy products. The objectives of this study were to establish simplified models to reflect the efficacy of the air frying process at varying temperatures and times on the quality attributes of falafel, and to optimize the frying conditions for producing air-fried falafel. Moisture content, color, fat content, hardness, and sensory evaluation of the fried falafel were analyzed under varied temperatures (140 °C, 170 °C, and 200 °C) and time periods (5 min, 10 min, and 15 min). Statistical analysis was then applied to obtain the best fit model that can describe the properties of fried falafel. Results indicated that moisture content, fat content, and L*-value of air-fried falafel were adversely related to the frying temperature and time, but the hardness and ΔE of fried falafel were increased as the frying temperature and time increased. Moreover, an increase followed by a decrease was shown for the appearance, aroma, crispness, taste, and overall preference scores with the increase in frying temperature and time. The regression analysis showed that the proposed models could be properly used for predicting the properties of the fried falafel. In addition, the overlaid plots resulted in the optimum frying temperature of 178.8 °C and time of 11.1 min. Interestingly, the fat content of the air-fried falafel reduced by 45% at optimal frying conditions compared with that for the deep-fat fried one at 180 °C for 7 min (control). In comparison, the air-fried falafel was lower in fat content, higher in hardness with more acceptable appearance and crispness scores than deep-fat fried falafel. Such information could be beneficial to the manufacturers of the falafel to produce an optimal and healthy product.

Sweet pepperincludes several vitamins and is regarded as a great source of bioactive nutrients, such as carotenoids and phenolic compounds, for human growth and activities. This work aimed to investigate the effects of the soil addition of growth stimulants, namely, effective microorganisms (EM), compost tea, fulvic acid, and yeast extract, and foliar applications of seaweed extract, on the vegetative growth, enzyme activity, phytohormones content, chemical constituents of plant foliage, fruit yield, and fruit quality of sweet pepper plants (Capsicum annuum L. cv. Zidenka) growing under greenhouse conditions. The results showed that the tallest plant, largest leaf area/plant, and heaviest plant fresh and dry weights were recorded after combining a soil addition of yeast extract and foliar spray with seaweed extracts at 3 g/L in two growing seasons. The highest number of fruit/plant, fruit yield/m2, fruit values of vitamin C (VC), total sugars, total soluble solids (TSS), and carotenoids, along with the highest leaf of cytokines, P, K, Fe, and total carbohydrates values, were obtained using a soil addition of fulvic acid and spray with seaweed extract at 3 g/L in the two seasons of study. These treatments also provided the lowest abscisic acid, peroxidase, and super oxidase dismutase values in the same conditions. Sweet pepper plants supplemented with compost tea and seaweed extract foliar spray at 3 g/L were the most promising for inducing the highest values of fruit fresh and dry weights, fruit length and diameter, and the leavesrichest in N, Zn, and Mn; inversely, it induced the lowest catalase levels in both seasons. The applications of EM, yeast extract, and seaweed extract could be applied for high growth, mineral levels, enzymatic activity, fruit yield, and nutritional value of sweet pepper fruit and minimizing environmental pollution.

N-acetylcysteine (NAC) and melatonin were reported to exert protective effects on testicular tissues. Thus, this study aimed to determine which of these is more efficient against obesity-induced testicular dysfunction in albino rats. A total of 32 adult male rats (195 ± 10 g) were divided into four groups: control, obese rats fed a high-fat diet (HFD), HFD+NAC (150 mg/kg per day, i.p.) and HFD+melatonin (10 mg/kg per day, i.p.), for 5 weeks. Testes and epididymis were weighed. Lipid profile, pituitary-testicular hormones, tumor necrosis factor α (TNFα), epididymal sperm parameters, testicular oxidant-antioxidant system, testicular and the epididymal histopathology and immunohistochemical localization for androgen receptors (AR) and Bax reaction were analyzed. Administration of NAC or melatonin significantly improved the lipid parameters, gonadal hormones, TNFα level, sperm count and abnormal morphology, oxidant-antioxidant system and the absolute testicular and epididymal mass with an enhancement of testicular architecture, AR expression and apoptosis as compared with that in the obese group. Additionally, as compared with the NAC group, the melatonin group had significantly reduced body mass index, total cholesterol, triglyceride, and TNFα and increased testosterone, sperm count, motility, superoxide dismutase activity, mitigated histomorphometrical changes, Bax expression, and increased testicular AR expression. Therefore, melatonin was more efficient than NAC in affording fortification against HFD-induced testicular dysfunction.

Purpose: Microsporidium is a spore-forming intracellular parasite that affects a wide range of hosts including humans. The tumor necrosis factor alpha (TNF-α) plays a key role in the immunity to infection with microsporidia. Recently, the TNF-α antagonists have proven successful in treating variable autoimmune diseases. In the current study, we aimed to investigate the impact of using TNF-α antagonists as a therapeutic regimen in the prevalence of infections with microsporidia. Materials and Methods: Diarrheal patients with distinct autoimmune diseases (n = 100) were assigned to the study. Patients taking anti-TNF-α medications (n = 60) were allocated to Group 1A and those undergoing non-TNF-α inhibitor treatment (n = 40) to Group 1B. Furthermore, patients with diarrhea without autoimmune disorders (n = 20) were allocated as controls. Stool specimens, 3 per patient, were collected and microscopically examined for microsporidia spores. A microsporidia-specific stool polymerase chain reaction was used to confirm the microscopic findings. Results: Microsporidia infection was identified in 28.3% (17/60), 10% (4/40), and in 5% (1/20) of patients in Group 1A, Group 1B, and in the control group, respectively. Overall, infection was significantly high in cases compared to the controls and in patients receiving TNF-α antagonists compared to patients not given TNF-α inhibitors (P < 0.05). Finally, infection was significantly higher in cases treated with TNF-α antagonists for ≥2 months compared to cases treated for <2 months of duration (P < 0.05). Conclusion: There was a significant increase in microsporidia infection in autoimmune disease patients undergoing treatment with TNF-α antagonists, and the duration of treatment is one of the risk factors. The study highlights the importance of microsporidia testing in immunocompromised patients, particularly those undergoing treatment with anti-TNF-α drugs and emphasises the need for awareness among clinicians regarding this opportunistic parasite.

Background: Ovarian cancer is the most common gynecological malignancy. In patients with advanced ovarian cancer, some biological parameters have prognostic implementations. P27kip1 is an inhibitor of a cycline-dependent kinase, its loss, can contribute to tumor progression. Objective: This study aimed to examine the importance of P27KIP1 protein in predicting the prognosis and response to neoadjuvant chemotherapy in patients with advanced ovarian epithelial cancer and to compare the outcomes of immunohistochemistry with Quantitative Real-time PCR. Patients and methods: We have studied P27KIP1expression by both immunohistochemistry and Quantitative Realtime PCR from 88 patients with advanced ovarian carcinomas undergone radical debulking surgery and received Paclitaxel followed by Cisplatin every 3 weeks for a total of 6 cycles. We also studied their association with both chemotherapy response and patient survival. Results: Nuclear expression of p27KIP1 protein was intense in 86 normal ovarian tissues and 42 of 88 carcinomas. The P27kip1mRNA expression level by qRT-PCR was very low in ovarian cancer tissues relative to its adjacent normal tissues. The results were statistically significant by both methods of determination. p27KIP1 expression was significantly related to good prognostic parameters as low stage tumors, differentiated tumors, absence of ascites, residual disease < 2 cm, and response to chemotherapy but not with histopathological type in case of determination by immunohistochemistry. Comparison of P27kip1 by both immunohistochemistry and qRTPCR with different prognostic parameters revealed no significant difference between both methods in the assessment of these parameters. In 4 years of follow-up, 20.5% of patients were alive without evidence of disease. 6.8% were alive with disease. The disease-related four -year survival rate for the whole group was 28.2%. In multivariate analysis, residual disease, histological type, tumor differentiation, ascites was of independent prognostic significance. Conclusion: In ovarian cancer, patients with loss of p27KIP1 expression are at a greater likelihood of disease progression, p27KIP1 may be used as a molecular marker to predict response to chemotherapy and prognosis. Both immunohistochemistry and qRT-PCR have equal reliability in the determination of p27 KIP1

Toxoplasma gondii (T. gondii), an intracellular parasite, establishes a chronic infection by forming cysts preferentially in the brain. TNF-α plays an important role in controlling the infection caused by this protozoan. Thus, the blockade of TNF-α could cause reactivation of latent toxoplasmosis infection as well as increase the risk of acute toxoplasmosis. This study evaluated the effect of etanercept, a TNF-α antagonist in reactivation of latent toxoplasmosis compared to the therapeutic effect of sulfadiazine and pyrimethamine in combination on the progress of the disease. A total of 40 laboratory-bred Swiss albino mice were infected with Me49 strain of T. gondii and divided into four groups: infected control group; treated group with sulfadiazine and pyrimethamine; treated group with etanercept and treated group with both etanercept and sulfadiazine and pyrimethamine. The mean number and size of tissue cysts in brain smears of mice of each group were determined and also, serum levels of TNF-α were assessed in different study groups by an enzyme linked immunosorbent assay. The results showed that the mean TNF-α level was significantly different in the treated groups compared to that in infected control group. The highest level of TNF-α was found in the infected controls. After treatment with etanercept alone or combined with sulfadiazine and pyrimethamine, it was significantly decreased. In this study, reactivation of latent toxoplasmosis was observed by a significant increase in the mean number and sizes of Toxoplasma tissue cysts in brains of mice with established chronic toxoplasmosis after treatment with etanercept alone or combined with conventional treatment compared to both untreated chronically infected controls and infected mice treated with sulfadiazine and pyrimethamine. It was concluded that etanercept, a TNF-α antagonist may play a role in reactivation of latent toxoplasmosis. So, serological screening for toxoplasmosis might offer a valuable aid for patients treated with this drug.

Strongyloides species is a helminth of worldwide distribution primarily in tropical and subtropical regions. It is the only soil-transmitted helminth with the ability for autoinfection so; it may lead to severe systemic manifestations especially in immunosuppressed patients. Chemotherapy is currently considered the best therapeutic option for strongyloidiasis but some drugs are expensive and others have side effects as nausea, diarrhea and headache. Strongyloides larva is resistant to most chemical agents so, search for plant extracts may provide other effective but less expensive treatment. Lawsonia inermis Linn, popularly known as Henna, has been proven to have antihelminthic, antibacterial and antifungal properties. The current study was carried out to evaluate the efficacy of Lawsonia inermis on Strongyloides spp. In vitro using scanning electron microscopy. Fifty Strongyloides species. larvae and free living females were incubated with different concentrations of Lawsonia (1, 10, 100 mg/ml), for different incubation periods (24, 48, 72 and 96 h) in comparison to the same concentrations of flubendazole at the same different time points. The results showed that Lawsonia inermis in a concentration of 10 mg/ml incubated with Strongyloides spp. female for 24 h affected the parasite cuticular surface in the form of transverse and longitudinal fissures and transverse depression in comparison to no cuticular change with flubendazole (100 mg/ml). This suggests that Lawsonia inermis may be a promising phytotherapeutic agent for strongyloidiasis.

Patients with chronic liver diseases (CLD) are often highly susceptible to parasitic infection due to a depressed immune system. The objective of this study was to detect the most commonly intestinal parasites found among Egyptian children with CLD. The present study was conducted on 50 children with CLD of different etiology (25 were having different intestinal symptoms, 25 without intestinal symptoms) and 50 non-CLD children with gastrointestinal complaints served as controls. All cases were subjected to stool examination and investigated by liver function tests. Also, anthropometric measurements were taken for all children including weight and height. It was found that the most commonly intestinal protozoa identified in the patients with CLD in order of frequency were: Entamoeba histolytica/Entamoeba dispar (16 %), Giardia lamblia (14 %), Blastocystis hominis (14 %), Cryptosporidium parvum (10 %), E. histolytica and G. lamblia (2 %), E. histolytica and B. hominis (2 %), G. lamblia and B. hominis (2 %), B. hominis and Entamoeba coli (2 %), Microsporidium (2 %) and no cases were found infected with Strongyloides stercoralis. As compared to the controls, the observed incidence of these organisms in CLD patients was significantly higher (p < 0.045) as regards stool examination by unstained techniques while, there was no significant difference between both groups as regards stool examination by stained techniques (p < 0.478). In addition, this study showed that the weight and height of studied patients were affected by parasitic infection while, there was no significant correlation between parasitic infection and liver function tests. In conclusion, chronic liver diseases affect the immunity of the patients as shown in significant increase in the incidence of intestinal parasites in cases compared to controls.

Acanthamoeba castellanii causes amoebic keratitis which is a painful sight-threatening disease of the eyes. Its eradication is difficult because the amoebas encyst making it highly resistant to anti-amoebic drugs, but several medicinal plants have proven to be more effective than the usual therapy. This study aimed to evaluate the in vitro amoebicidal activity of ethanol extracts of Arachis hypogaea L. (peanut), Curcuma longa L. (turmeric), and Pancratium maritimum L. (sea daffodil) on A. castellanii cysts. Acanthamoeba were isolated from keratitic patients, cultivated on 1.5% non-nutrient agar, and then incubated with different concentrations of plant extracts which were further evaluated for their cysticidal activity. The results showed that all extracts had significant inhibitory effect on the multiplication of Acanthamoeba cysts as compared to the drug control (chlorhexidine) and non-treated control, and the inhibition was time and dose dependent. The ethanol extract of A. hypogaea had a remarkable cysticidal effect with minimal inhibitory concentration (MIC) of 100 mg/ml in all incubation periods, while the concentrations of 10 and 1 mg/ml were able to completely inhibit growth after 48 and 72 h, respectively. The concentrations 0.1 and 0.01 mg/ml failed to completely inhibit the cyst growth, but showed growth reduction by 64.4-82.6% in all incubation periods. C. longa had a MIC of 1 g and 100 mg/ml after 48 and 72 h, respectively, while the concentrations 10, 1, and 0.1 mg/ml caused growth reduction by 60-90.3% in all incubation periods. P. maritimum had a MIC of 200 mg/ml after 72 h, while the 20-, 2-, 0.2-, and 0.02-mg/ml concentrations showed growth reduction by 34-94.3% in all incubation periods. All extracts seemed to be more effective than chlorhexidine which caused only growth reduction by 55.3-80.2% in all incubation periods and failed to completely inhibit the cyst growth. In conclusion, ethanol extracts of A. hypogaea, C. longa, and P. maritimum could be considered a new natural agent against the Acanthamoeba cyst.
